The contraindications, foods, and restrictions of Rezdifra must be strictly followed to ensure medication safety.

1 Taboo population

(1) Pregnant women: Based on animal studies showing embryotoxicity, use should be avoided during pregnancy, and strict risk assessment should be conducted if necessary.

(2) Breastfeeding women: There is no data available on the distribution of breast milk. It is recommended to suspend breastfeeding during the treatment period and within one week after the last dose.

(3) Liver dysfunction: The safety of use in patients with severe liver dysfunction is not clear, and should be avoided or closely monitored.

2 Forbidden Foods

(1) Citrus products: may increase drug exposure by inhibiting CYP3A4 enzyme, leading to an increased risk of side effects.

(2) High fat diet: Taking it together with a high-fat meal may affect the drug absorption rate, and it is recommended to take it on an empty stomach at a fixed time.

3 Clinical monitoring requirements

(1) Liver function monitoring: transaminase should be checked before treatment, and monthly follow-up should be conducted during treatment. If ALT/AST>3 times the normal value, medication should be suspended.

(2) Electrocardiogram examination: QT interval should be regularly monitored before medication and during treatment. QTcF>450ms is contraindicated, and dose reduction is required if prolongation occurs.

4 Adverse Reaction Responses

(1) Photogenic reaction: Sunscreen with SPF ≥ 30 should be used during medication to avoid direct sunlight. If severe rash occurs, the medication should be discontinued.

(2) Gastrointestinal reactions: It is recommended to take after meals. If there is persistent vomiting/diarrhea, electrolytes should be supplemented and the dosage adjusted.

5 Restrictions

(1) Driving restrictions: Avoid driving or operating machinery during medication as it may cause dizziness or blurred vision.

(2) Sunscreen requirements: Strict sun protection is required during treatment to avoid photosensitive reactions caused by exposure to ultraviolet rays.

(3) Contraceptive requirements: Patients of childbearing age must take effective contraceptive measures during treatment and within 4 months after discontinuing medication.

(4) Combination therapy: It is prohibited to use potent CYP3A4 inhibitors (such as ketoconazole) in combination, and the dosage needs to be adjusted.
