The use of Ixazomib involves key elements such as dosage regimen, administration method, and treatment cycle, and must be strictly adjusted according to indications and individual patient conditions. 1. Standard usage and dosage

(1) The recommended dosage is 4mg orally, once a week (fixed administration day), taken continuously for 3 weeks and stopped for 1 week, forming a 28 day treatment cycle.

(2) Should be used in combination with 25mg of lenalidomide (Day 1-21) and 40mg of dexamethasone (Day 1/8/15/22) until disease progression or intolerable toxicity occurs.

2 Precautions for administration

(1) It should be taken on an empty stomach and administered at least 1 hour before or 2 hours after meals. The whole pill should not be chewed.

(2) If the missed dose is ≤ 72 hours, it can be immediately replenished. If it exceeds 72 hours, the dose will be skipped and cannot be doubled.

3 dose adjustment plan

(1) Liver dysfunction: For patients with moderate to severe damage, the dosage should be reduced to 3mg/week, and it is contraindicated for end-stage liver disease.

(2) Hematological toxicity: When the platelet count is less than 30 × 10 ⁹/L or the neutrophil count is less than 0.5 × 10 ⁹/L, the administration should be suspended and downgraded to 3mg/week after recovery.

4. Medication for special populations

(1) Elderly patients (≥ 65 years old) do not need to adjust the dosage, but adverse reactions need to be closely monitored.

(2) The safety of medication for children has not been established. It is prohibited during pregnancy and feeding should be stopped during lactation.
