Larotrectinib should be taken as a whole tablet, and the recommended dosage should be adjusted according to age and weight. It can be taken with food or on an empty stomach, and treatment should be continued until disease progression or intolerable toxicity occurs.

1. Administration method

(1) Oral administration, twice daily (approximately once every 12 hours).

(2) Swallow the whole piece, do not cut, crush or chew.

(3) It can be taken with food or on an empty stomach, and the medication interval should be consistent.

2 Recommended Dosage

(1) Adults and children (body surface area ≥ 1.0m ²): 100mg each time.

(2) Children (body surface area<1.0m ²): 100mg/m ² per session.

(3) The dosage needs to be adjusted according to adverse reactions, and can be reduced to a minimum of 50mg or 50mg/m ².

3 Omission Handling

(1) If missed and more than 6 hours before the next medication, it should be taken immediately.

(2) If the time until the next medication is ≤ 6 hours, skip the missed dose and do not double the dosage.

4. Medication for special populations

(1) Liver dysfunction: Mild cases do not require dose adjustment, moderate to severe cases require dose reduction to 75%.

(2) Renal insufficiency: Mild to moderate without adjustment, severe lacking data.

(3) Prohibited during pregnancy, medication should be discontinued or breastfeeding should be stopped during lactation.
