1. Standard usage and dosage

The recommended dose of Fruquintinib is 5mg once daily, taken continuously for 21 days and stopped for 7 days (28 days per cycle). It should be swallowed whole and can be taken with food or on an empty stomach, but avoid taking with grapefruit juice.

2. Dose adjustment plan

(1) Adjustment of liver dysfunction

Mild liver dysfunction (Child Pugh A grade) patients do not require dose adjustment; For patients with moderate to severe liver dysfunction (Child Pugh B/C grade), it is recommended to reduce the dosage to 4mg or 3mg per day.

(2) Adverse reaction adjustment

If grade 3 hypertension occurs, medication should be suspended until it returns to grade 1 or below, and subsequent treatment should be reduced by one dose level; Grade 4 hand foot syndrome requires permanent discontinuation of medication.

3. Medication for special populations

(1) Pregnant women

Based on animal studies showing teratogenicity, it is contraindicated during pregnancy. Before medication, it is necessary to confirm the pregnancy status, and effective contraceptive measures should be taken during the treatment period and 2 weeks after discontinuation of medication.

(2) Lactating women

There is currently no data on drug concentration in human breast milk. It is recommended to stop breastfeeding during the treatment period and within 2 weeks after the last dose.

4. Absolutely Forbidden Foods

(1) Grapefruit and its products

Grapefruit juice contains potent CYP3A inhibitors, which may increase the blood concentration of fruquintinib by more than twice, increasing the risk of adverse reactions. Beverages containing fresh juice, fruit pulp, and grapefruit ingredients should be avoided.

(2) Seville Orange Products

This variety of orange contains furan coumarin compounds similar to grapefruit, which may interfere with drug metabolism. Avoid consuming derivatives such as jam and citrus tea.
