Elafibrino should be strictly used under the guidance of a doctor, and correctly mastering the usage and dosage, understanding the efficacy evaluation indicators, and key precautions are the core to ensure safe and effective treatment. The following is divided into three parts for explanation.

1、 Usage and dosage

Regular dose: once daily, 80mg each time, taken at a fixed time each day, recommended to be taken with meals to increase absorption.

Dose adjustment: If the therapeutic effect does not meet expectations after 12 weeks of treatment, it can be increased to 120mg per day after evaluation by a doctor, but liver function needs to be monitored simultaneously.

Omission treatment: If you forget to take your medication on the same day, you can take it again within 12 hours after remembering it; If it is close to the next day's medication time, skip the missed dose and do not double the dose.

Special population: Patients with mild liver injury (Child Pugh A) do not require adjustment, but need to strengthen monitoring; Patients with severe liver dysfunction are prohibited from using it.

2、 Efficacy evaluation

Main outcome measures: After 12 weeks of treatment, re examination of transaminase (ALT/AST) levels showed a significant decrease, indicating effective treatment.

Secondary assessment: accompanied by symptom improvement, such as reduced discomfort in the upper right abdomen and alleviation of fatigue.

Long term follow-up: It is recommended to have a liver elasticity test or imaging examination every 6 months to evaluate the dynamic changes of fibrosis.

Efficacy evaluation node: If there is no improvement or further deterioration of liver function after 24 weeks of treatment, the necessity of treatment needs to be re evaluated.

3、 Precautions

Liver function monitoring: ALT, AST, GGT, and total bilirubin are tested monthly before and after medication. If ALT rises more than three times the upper limit of normal, medication should be suspended.

Attention to renal function: Check blood creatinine and estimate glomerular filtration rate every 3 months. If the increase in creatinine exceeds the baseline value by 30%, the medication should be reduced or stopped.

Drug interactions: Avoid combination with potent CYP3A4 inducers (such as rifampicin) and OATP inhibitors (such as cyclosporine) to avoid affecting efficacy or increasing toxicity.

Weight management: During medication, regular monitoring of weight is necessary. If there is unexplained weight gain, dietary and exercise plans should be adjusted.

Taboo reminder: Pregnant, lactating, and biliary obstruction patients are prohibited from using it.

All medication decisions must be made under the guidance of a doctor. If there is any obvious discomfort or abnormal laboratory indicators during the treatment period, timely medical attention should be sought. Do not increase or decrease the dosage or stop medication without authorization.
