Precautions for taking Relugole: QT/QTc interval prolongation, hepatotoxicity, and drug interactions

Rulugoli reduces the secretion of luteinizing hormone (LH) and follicle stimulating hormone (FSH) by blocking the GnRH receptor in the anterior pituitary gland, thereby lowering estrogen levels in the body.

1、 QT/QTc interval prolongation

High risk populations: those with congenital long QT syndrome, electrolyte disorders (low potassium, low magnesium), heart failure, and bradycardia should use with caution.

Avoid co administration: Do not use with drugs known to prolong QTc interval (such as amiodarone, sotalol, moxifloxacin); If combined use is necessary, the frequency of electrocardiogram monitoring should be increased.

Electrocardiogram monitoring: Regularly review the electrocardiogram before and during treatment, especially when there are signs of dizziness, palpitations, or fainting.

2、 Hepatotoxicity

Liver function examination: Before taking medication and during treatment, transaminase and bilirubin should be regularly checked. If there is fatigue, nausea, jaundice, or discomfort in the upper right abdomen, timely follow-up is necessary.

Dose adjustment or discontinuation: If transaminase elevation exceeds the normal upper limit by 5 times or is accompanied by bilirubin elevation, medication should be suspended and the cause evaluated.

Basic liver disease: Individuals with a history of active liver disease or liver injury should undergo a thorough risk benefit assessment before use.

3、 Drug interactions

P-gp/BCRP substrate drugs (such as digoxin and dabigatran etexil): When used in combination, the exposure to these drugs can be increased, and the efficacy and toxicity need to be monitored.

Strong P-gp inducers (such as rifampicin and carbamazepine): can significantly reduce the blood concentration of rilugole, and should be avoided in combination; If unavoidable, medication should be taken intermittently or the plan adjusted.

Antacids/cationic binders: Take at least 2 hours apart from preparations containing calcium, magnesium, and aluminum to avoid affecting absorption.
